Genetic engineering and modern enzymology

Abstract: The role of genetic engineering in the solution of problems involving enzymology is demonstrated. The method for the cloning and mutagenesis of protein and enzyme genes and the possibilities of genetic engineering for the study of the structure and functions of enzymes and their active centres and for the determination of the relation between the structure and stability of protein molecules are examined. The role of directed site-specific mutagenesis in the study of the structure and mechanism of the action of enzymes as well as their stability is indicated. The investigation of the mechanism and factors determining the stability of enzymes with the aid of aminoacid substitutions has led to prospects for protein design – the creation of proteins with new specified properties and improved stability.
