Inhibitors of topoisomerases I and II: chemical structure, mechanisms of action and the role in cancer chemotherapy

Abstract: The modern literature data on topoisomerase I and II inhibitors as potential antitumour drugs are integrated and analyzed. The functions and mechanisms of action of topoisomerases and the molecular mechanisms of interaction of low molecular weight compounds with topoisomerases are considered. The topoisomerase inhibitors of a variety of chemical classes are described systematically. The tests used to elucidate the topoisomerase inhibition and the potential of computer-assisted modelling for the rational design of new drugs for cancer chemotherapy are presented.
